The main focus of our group is to new chromatin factors important for maintaining genome stability. With this, we aim to uncover novel epigenetic mechanisms that control chromosome stability and genetic transmission, of which are pertinent to cell growth, tissue differentiation and embryo development.
Our research aim is to uncover fundamental epigenetic mechanisms that regulate transcriptional silencing at repetitive DNA sequences in the genome. Histone variant H3.3 and its chaperone ATRX (Alpha Thalassemia Mental Retardation X-linked) are two key chromatin regulators important for maintaining telomere repeat DNA stability and function. Recent studies have shown that both H3.3 and ATRX are frequently mutated in human cancers, including the brain and bone cancers. We use CRISPR CAS9 gene editing system, highly advanced cellular imaging and high throughput DNA and RNA sequencing to investigate the genome-wide epigenetic defects and DNA instability associated with H3.3 and ATRX mutations in cancers.
