Hybridoma Sub-cloning
MADP staff set up 2 Terasaki plates (https://shop.gbo.com/en/row/products/bioscience/immunology-hla/terasaki-plates/653180.html) per cell line using single cell deposition in FlowCore, followed by visual inspection of the wells to confirm that only 1 cell is present. Each well that passes the “1 cell” test is then grown up to a culture to be tested by ELISA. The highest 2 cell lines are retained and, for the top line, the whole process is repeated for a second time to generate a monoclonal cell line. The second cell line is frozen as a back-up cell line.
For the final monoclonal cell line we freeze 2 vials of cells that can either be stored in MADP or sent to the customer.
